or do I have first upgrade to Lightroom 3.x (cheap right now) and then update to 4.0 ?
The Lightroom 4.0 beta looks (and is) really cool so I would like to update.

In summary, it says: Databases would be upgradable, but Adobe doesn't announce the upgrade path(s) and pricing until the product is officially announced.

Your question is understandable due to all the sales right now. If I were Adobe, rather than deny 2 to 4 upgrades, I'd just charge more if I wanted to be a little mean about it, but I wouldn't charge more than what the current sale prices are for LR3 added on to the price of the 3 to 4 version upgrade price, and probably would make it a little cheaper than that to reward customer loyalty.

Having said that, Adobe has been trying to optimize their revenue with subscriptions and other innovations to their traditional upgrade paths, so perhaps they have turned their imagination to the LR upgrades as well. The bottom line is that no one knows who might speculate, and no one can say if they know.

I can't imagine Adobe would have a versioned schema that was not upgradeable. It would serve no useful purpose.

One favorable indication is that an upgrade from Lightroom 1 directly to Lightroom 3 was (and still is) permitted:
