First, I really like being able to change values in just shadow or midtone or highlights in the Beta version of LR4. I hope that feature makes it into PS and PSE.

I would really love to see Focus Stacking image merging in one of more of LR, PS or PSE. It would be great to have both HDR Pro and Focus Stacking in Photoshop CS.

I would also like to see HDR exposure control and focus control for focus stacking added to the tether function.

I taught photo imaging for the USAF photo mapping and reconnaissance and I am new to digital imaging and digital photo editing. I am now learning LR3&4, PSE10 and PS CS5.1 as a registered user.

I carried a Deardorff 8X10 and 4X5 field camera for landscape and nature photography when I was a young man and used Zone System extensively. My dark room is now used for storage and I am excited about the possibilities of digital imaging and editing.

Photoshop already has a focus stacking feature (and more if you have Extended).
Image -> Auto Blend -> Stack Images

Photoshop also has more extensive value and color controls and masking than Lightroom.
